Abstract. Djoemharsjah IS, Ambarwati R, Rahayu DA. 2023. DNA barcoding Clithonsp. (Gastropoda: Neritidae) from Badur Beach, Madura, Indonesia, based on COI gene molecular marker. Biodiversitas 24: xxxx. Neritidae is polymorphic with various shell colors and patterns; therefore, they were doubts about the identifying species genus Clithon, namely Clithon sp. Beach of Madura Island, Sumenep District, East Java Province, Indonesia. In addition, could provide genetic information using short sequences to quickly precisely identify species. This study aimed Cytochrome Oxidase subunit I(COI) genes analysis phylogenetic relationships. The research methods include sampling, sample preservation, morphological identification species, isolation, amplification, electrophoresis, sequencing Sanger method bioinformatics software. results barcode obtained a sequence length 490bp similarity value three samples between 96.75 98.97%. Barcode Life Data System consisted variations nucleotide bases, average distance in-group was 1.74% as Phylogenetic tree in same clade sowerbianum Récluz 1843 mertonianum Neighbor-Joining Tree Maximum Likelihood bootstrap values 96-100. Therefore, markers successfully identified Indonesia C. sowerbianum.

Molecular investigation and DNA Barcoding of Platycephalus indicus from the Persian Gulf
This study aimed to use mitochondrial DNA barcoding method to undrestand better toxanomic status of the Platycephalus indus and find genetic linkage with other reported specimens from different parts of the world as well as to manage the optimal utilization of native species and sustainable conservation. Sampling was performed from coastal waters of Hormozgan province. DNA was extracted from th...
متن کاملMolecular identification of the genus Thryssa based on DNA barcoding.
DNA barcoding is an effective method for identifying species by analyzing one or a few short standardized DNA sequences. In this study, we examined the utility of mitochondrial cytochrome oxidase subunit I (COI) sequences as a DNA barcode for the identification of six species belonging to the genus Thryssa: T. dussumieri, T. hamiltonii, T. kammalensis, T. mystax, T. setirostris, and T. vitriros...
متن کاملLimitations of cytochrome oxidase I for the barcoding of Neritidae (Mollusca: Gastropoda) as revealed by Bayesian analysis.
The mitochondrial DNA (mtDNA) cytochrome oxidase I (COI) gene has been universally and successfully utilized as a barcoding gene, mainly because it can be amplified easily, applied across a wide range of taxa, and results can be obtained cheaply and quickly. However, in rare cases, the gene can fail to distinguish between species, particularly when exposed to highly sensitive methods of data an...
متن کاملWith a little help from DNA barcoding: investigating the diversity of Gastropoda from the Portuguese coast
The Gastropoda is one of the best studied classes of marine invertebrates. Yet, most species have been delimited based on morphology only. The application of DNA barcodes has shown to be greatly useful to help delimiting species. Therefore, sequences of the cytochrome c oxidase I gene from 108 specimens of 34 morpho-species were used to investigate the molecular diversity within the gastropods ...
متن کاملmolecular phylogeny of malaysian pangasiid based on cytochrome c oxidase i (coi) dna sequences
the molecular phylogenetic trees of 42 representative samples of malaysian pangasiid were constructed using cytochrome c oxidase subunit i (coi). the topologies resulting from molecular analyses, maximum parsimony, maximum likelihood, and bayesian inference, revealed the paraphyly of pangasius genus and monophyly of p. nasutus, p. bocourti, p. micronemus, and p. hypopthalmus with high support v...
